目的观察mTOR及其信号通路分子在食管鳞状细胞癌和外周T淋巴细胞的表达。方法收集40例食管鳞状细胞癌术后癌组织、癌旁组织和外周T淋巴细胞,运用免疫组化、实时定量聚合酶链反应及流式细胞术检测mTOR、raptor及p-TSC2的表达量。结果癌组织mTOR和raptor表达量明显高于癌旁组织。临床Ⅲ~Ⅳ期mTOR蛋白的阳性表达率明显高于临床Ⅰ~Ⅱ期。患者外周血T淋巴细胞p-mTOR、p-TSC2的表达量高于正常人。结论食管鳞状细胞癌及外周T淋巴细胞中mTOR信号通路活化程度强于正常人。
To evaluate the expression of mTOR and its signal pathway in esophageal squamous cell carcinoma tissues and peripheral T lymphocytes,40 cases of esophageal squamous cell carcinoma tissues,tumor-adjacent tissues and peripheral T lymphocytes were collected.Immunohistochemistry,real-time PCR and FACS were employed to detect the expression levels of mTOR,raptor and p-TSC2.Results showed that the expressions of mTOR and raptor in esophageal squamous cell carcinoma tissues were significantly higher than those in tumoradjacent tissues,and the positive expression rate of mTOR in Ⅲ-Ⅳ phase patient were significantly higher than that inⅠ-Ⅱ phase patient.Furthermore,the expressions of p-mTOR and p-TSC2 in peripheral T lymphocytes of patients were significantly higher than those in health.In conclusion,the signal pathway of mTOR is much activated in esophageal squamous cell carcinoma tissues and peripheral T lymphocytes.
