目的研究高浓度葡萄糖对体外培养兔角膜上皮细胞增殖活性的影响,建立糖尿病性兔角膜上皮细胞培养模型。方法按照析因设计方法将葡萄糖浓度(25、40、60 mmol/L)和培养时间(24、48、72 h)进行全面组合,应用SunBioTM Am-Blue细胞增殖与活性检测试剂检测兔角膜上皮细胞增殖活性,倒置显微镜观察细胞形态。结果高浓度葡萄糖对兔角膜上皮细胞增殖活性的影响有统计学意义(F=276.318,P〈0.01),作用时间对细胞增殖活性的影响有统计学意义(F=38.727,P〈0.01),葡萄糖浓度和作用时间之间有交互作用(F=2.687,P〈0.05)。结论高浓度葡萄糖对体外培养兔角膜上皮细胞增殖活性有抑制作用,随时间的延长抑制作用加强。
Objective To study the effects of high glucose on proliferation activity of cultured rabbit corneal epithelial cells in vitro,and to establish culture model of diabetic rabbit corneal epithelial cells.Methods Rabbit corneal epithelia cells were cultured in glucose concentrations at 25,40 and 60 mmol/L,for 24,48 and 72 hours apiece according to factorial design.SunBioTM Am-Blue cells proliferation and activity assay kit was applied to detect proliferation activity of cultured rabbit corneal epithelial cell in vitro;and microplate reader was used for assessing the OD values,which was the evaluation index of proliferation activity of cell.Cell morphology was observed by inverted microscope.Experiment was repeated three times. Results Significant inhibitation was observed in cell proliferation of cells cultured with high glucose(F=276.318,P0.01),with significant differences among cell with different culture time(F=38.727,P0.01).Significant interaction was observed between glucose concentration and culture time on cell proliferation(F=2.687,P0.05). Conclusion High glucose inhibits the proliferative activity of cultured rabbit corneal epithelial cells in vitro in a time-dependent manner.
