目的探讨丹参酮IIA(tanshinonelIA)对肢体缺血再灌注大鼠血液流雯学的影响。方法24只Wistar大鼠随机分为正常对照组(control组)、再灌注组(IR组)和丹参酮IIA治疗组(DS-201组)。以止血带法制作大鼠肢体缺血/再灌注模型,DS-201组在恢复血流再灌注前30min静脉推注丹参酮IIA磺酸钠(简称DS-201)注射液(25mg/kg),而前两组推注等体积25%葡萄糖注射液。测定血液SOD、MDA及血液流变学指标。结果与对照组相比,IR组血MDA明显升高,SOD活性降低,全血黏度、红细胞聚集指数、红细胞压积明显增大,红细胞变形指数减小(P〈0.05);与IR组比较,DS.201组血MDA降低(P〈0.05),SOD显著升高(P〈0.05),大鼠全血黏度(P〈0.05)、红细胞聚集指数(P〈0.05)降低,红细胞变性指数(P〈0.05)升高。结论丹参酮IIA可改善再灌注大鼠的血液流变性,其作用可能与提高大鼠抗氧化能力有关。
Objective To observe the effects oftanshinoneIIA on hemorheology of rats that suffured from limb ischemia-reperfusion. Methods 24 rats were divided into control group, ischemia reperfusion group and DS-201 group at random. Ischemia reperfusion model was reproduced as following, the model rats underwent 4 hours ischemia and 4 hours reperfusion of hind limbs. 30 minutes before reperfusion, the tanshinoneIIA was injected into animals in group DS-201(25mg/kg)o the former two groups were injected the same volume of 25% glucose injection. Measured the activity of superoxide dismutase(SOD) and the levels of malondialdehyde (MDA) in plasma. The changes of hemorheology in different groups were detected respectively, Results Compared with the control group, IR group was significantly higher serum MDA, SOD activity decreased blood viscosity, erythrocyte aggregation index, significantly increased hematocrit, erythrocyte deformation index reduced (P〈0.05); compared with IR group, DS -201 MDA lower blood group (P〈0.05), SOD increased significantly (P〈0.05), rat whole blood viscosity (P〈0.05), erythrocyte aggregation index (P〈0.05) reduced red blood cell degeneration index (P〈0.05) increased.. Conclusion TanshinoneIIA can improve hemorheology in reperfusion rat, the effect may be related to improve the antioxidant ability of rats.
