背景:骨性关节炎的防治是目前骨组织工程的的研究热点。目的:在灵长类动物建立膝关节骨性关节炎模型,并对建立的模型进行评估。方法:将食蟹猴麻醉后,切断左膝关节前后交叉韧带,并同时切除内侧半月板,而另一侧膝关节不予处理。建模后常规给予抗生素预防感染。建模后第4,8,12周观察猴关节活动情况、软骨Mankin评分、软骨组织病理切片等方面进行评估。结果与结论:建模后食蟹猴饮食正常,活动量逐渐增加,建模后第4周关节表面可见软骨有轻微磨损,第8周软骨的颜色变暗,磨损更严重,第12周软骨缺损,且可见轻微骨质增生。病理切片见软骨组织间纤维组织增生,软骨细胞散在分布,连续性中断,软骨细胞密度不均,部分区域软骨细胞增生。结果证实,将食蟹猴半月板切除,切断交叉韧带,并允许其自由活动能够成功制备骨性关节炎的模型,为以后的骨性关节炎治疗研究奠定基础。
Background: Current research focus on osteoarthritis prevention and treatment. Objective: To establish osteoarthritis model of primate and to evaluate whether the model successfully. Method: We anaesthetized the monkey firstly, and then transected the anterior cruciate ligament and removed the medial meniscus. The other knee just opened articular cartilage,closed the articular cartilage immediately. To prevent infection routine postoperative administration of antibiotics is necessary. After operative, we evaluated the model through the following aspects: joint activities, Mankin's score, pathological section of cartilage. Results and Conclusion: The monkey increased the amount of activity gradually. In the forth week,Cartilage has slight wearon the articular surface. In the eighth week, we found the dull articular cartilage in the operation knee. In the twelfth week, the thickness of cartilage is thinner than contrast group, and to be able to see the bone hyperpiasia. The method of the anterior cruciate ligament transection and the medial meniscus removal is successful to establish the osteoarthritis of monkey. The significance of this experiment was to lay the foundation for the follow up prevention and cure of osteoarthritis.
