目的同时测定山楂叶提取物中2″-O-牡荆素葡萄糖苷(Glv)、2″-O-牡荆素鼠李糖苷(Rhv)、芦丁(Rut)、牡荆素(Vit)和金丝桃苷(Hps)的含量。方法采用HPLC法,色谱柱为DiamonsilTMC18柱(200 mm×4.6 mm,5μm),流动相A为甲醇,B为乙腈-四氢呋喃(1:19),C为0.5%冰醋酸,梯度洗脱。0-15 min,流动相A由1%-21%,B由19%-21%;15-17 min,A由21%-65%,B保持5%;17-20 min,A由65%-1%,B由5%-19%;流速1.0 ml·min^-1;检测波长360 nm;柱温30℃。结果Glv、Rhv、Rut、Vit、Hps的平均加样回收率分别为99.72%、100.27%、99.48%、99.31%和100.83%,其RSD分别为1.26%、1.69%、1.61%、1.70%和1.31%(n=6)。结论所建方法简便、快速、准确、可靠,可用于山楂叶提取物的质量控制。
OBJECTIVE To establish an HPLC method to determine simultaneouly 2" - O - Glucosylvitexin ( GLV), 2" - O - Rhamnosylvitexin (RHV), Rutin (RUT), Vitexin (VIT) and Hyperoside (HP) in Hawthorn leaves extract. METHODS The DiamonsilTM C18 column (200 mm × 4. 6 mm, 5 μm) was used at the temperature of 30 ℃ ; the mobile phase consisted of A: CH3OH, B : CH3CN - THF( 1 : 19), C : 0.5% HAc ; gradient elution profile was 0 - 15 min,A: 1% - 21%, B : 19% - 21% , 15 - 17 min, A : 21% - 65%, Bkeep5%; 17 -20min, A :65% -1%, B:5% -19%. Theflowratewas1.0ml·min^-1. The UV detector was set at 360 nm. RESULTS The average recoveries of GLV, RHV, RUT, VIT and HP were 99. 72%, 100.27%, 99.48% , 99.31% and 100.83% , with RSD of 1.26%, 1.69% , 1.61% , 1.70% and 1.31% (n = 6), respectively. CONCLUSION The method is simple, rapid, accurate and reliable. It can be used to control quality of Hawthorn Leaves Extract and its preparations.
