目的 研究阳荷花苞的降血糖活性及所含的化学成分。方法 将阳荷花苞粗提物作用于胰岛素抵抗的Hep G2细胞,用葡萄糖试剂盒检测其降血糖活性,MTT法检测细胞活力,然后通过反复的硅胶柱层析、Sephadex LH-20柱色谱及重结晶分离、纯化阳荷花苞粗提物,通过理化常数和波谱分析鉴定化合物的结构。结果 阳荷花苞粗提物具有显著的降血糖效果,且细胞毒性较小。并从中分离鉴定了7个化合物,分别为:β-谷甾醇(1)、胡萝卜苷(2)、胸腺嘧啶(3)、尿嘧啶(4)、对羟基苯甲酸(5)、3-羟基-4-甲氧基苯甲酸(6)、蔗糖(7)。其中化合物3,4,5,6为首次从阳荷花苞中分离得到,且化合物3和4为首次从姜科植物中分离得到。结论 阳荷花苞降血糖作用显著,有必要对这种药食同源的植物进行研究并开发利用。
Objective To study the hypoglycemic activity of the extract from the flower bud of Zingiber striolatum Diels and its chemical constituents. Methods The insulin-resistant HepG2 cells were treated with the crude extract from the flower bud of Zingiber striolatum Diels. The hypoglycemic activity of the extract was measured with glucose kit, the cell viability was detected by MTT method. The chemical constituents were isolated and purified by repeated silica gel column chromatography, Sephadex LH-20 column chromatography and recrystallization. Their structures were identified by the analysis of the physiochemical property and spectral data. Results The ethanol extract from the flower bud ofZingiber striolaturn Diels showed significant hypoglycemic activity and less cytotoxicity in HepG2 cells. Seven compounds were obtained from the extract by repeated column chromatography and recrystallization. These compounds were identified as follows: daucosterol (1), sitosterol (2), thymine (3), uracil (4), p-hydroxybenzoic acid (5), 3-hydroxy-4- methoxybenzoicacid (6), D(+)-sucrose (7). Compound 3, 4, 5, 6 were isolated from the flower bud of Zingiber striolatum Diels for the first time, what is more, compound 3 and 4 were isolated from Zingiberaceae for the first time. Conclusion The extract from the flower bud of Zingiber striolatum Diels has significant hypoglycemic activity, further research and exploitation is a must to maximize the application of it.
