目的:观察丁酸钠(NaB)对体外培养的胃癌细胞系BGC823的生长抑制作用,并进一步探讨其作用机制。方法:应用MTT法检测细胞增殖活性;流式细胞仪检测细胞周期分布;Annexin V—FITC染色法检测细胞凋亡率。结果:不同浓度NaB处理胃癌细胞BGC82324~96h后.细胞增殖显著受到抑制,并呈浓度(P〈0.01)及时间(P〈0.05)依赖性;1.0,3.0,5.0mmol/LNaB处理72h后,BGC823细胞G0/G1期细胞数量显著增加,S期细胞数量显著降低(P〈0.01);各浓度组细胞凋亡率均显著增加(P〈0.01)。结论:NaB对胃癌细胞系BGC823具有生长抑制作用,其机制与阻滞细胞周期及诱导凋亡有关。
Objective: To investigate the effect of sodium butyrate (NaB), one of histone deacetylase inhibitors (HDACI), on the proliferation of human gastric cancer cell line BGC823 and its mechanism, Methods: BGC823 cells cultured in RPMI 1640 were treated with 0.5, 1. 0, 3. 0, and 5.0 mmol/L of NaB. MTT assay, flow cytometry, and Annexin V-FITC staining were performed to determine the cell proliferation activity, cell cycle, and apoptotic rate. Results: After BGC823 cells were treated with different concentrations of NaB for 24 to 96 hours, the cell proliferation activity was inhibited in a dose- and time-dependent manner ( P 〈 0.01, P 〈 0.05 ). The number of cells in G0/G1 phase significantly increased and the number of cells in S phase significantly decreased after the cells were treated with 1.0, 3.0, and 5.0 mmol/L of NaB for 72 hours (both P 〈 0.01 ). The apoptotic rate significantly increased in cells treated with different concentrations of NaB (all P 〈0.01 ). Conclusion: NaB could inhibit the growth of human gastric cancer cell line BGC823 by blocking cell cycle and inducing apoptosis.
