目的:初步探讨静电纺人工神经移植物丝素蛋白-PLGA神经导管(electrospun silk fibroin-PLGA nerve conduits,SP-NGCS)的生物相容性。方法:(1)体外实验:SP-NGCS与施万细胞共培养,形态观察与MTT检测。(2)体内试验:将SP-NGCS植入大白兔皮下,术后2W,4W,8W,12W,24W取材,切片后观察局部组织反应。结果:施万细胞生长状态良好,无毒性反应,且肉眼观察和形态组织学染色观察,组织炎症反应轻微,SP-NGCS在24W时大部分降解。结论:SP-NGCS生物相容性良好,是理想的人工神经移植物。
Objective:To evaluate the biocompatibility of the electrospun Silk fibroin/PLGA nerve conduits in vitro and in vivo. Methods: In vitro: Schwann cells were implanted in the extraction of the SP NGCS and the morphology was observed and MTT analysis was performed. In vivo: SP-NGCS were subderma implanted ,harvested at 2,4,8,12,24 weeks after operation and processed for histologie analysis. Results: SP-NGCS are beneficial to the survival of Schwann cells without exerting any significant eytotoxic effects and there was mild inflammatory reaction at the implant sites. NGCS were almost absorbed after 24 weeks. Conclusion: SP-NGCS have an excellent compatibility and can be used as nerve grafts.
