目的:探讨柴芩承气汤(CQCQD)对急性坏死性胰腺炎大鼠胰腺组织抗生素浓度的影响。方法:18只SD大鼠随机分成三组(正常组、ANP组、CQCQD组),每组6只。ANP造模采用3.5%牛磺胆酸钠(1ml/kg)逆胰胆管注射。造模成功后6h,大鼠腹腔注射CTX(420mg/kg,Q12h.连续72h);CQCQD组同时管喂中药(20ml/kg,Q8h,连续72h)。于第72h最后一次注射CTX15min后取血样和胰腺组织。用高效液相色谱法测量血清及胰腺组织的CTX浓度。结果:大鼠血清CqX浓度在三组间无差异。与正常组比较,ANP组大鼠胰腺组织的CTX浓度由前者的18.6±1.7(μg/ml)降至4.4±0.6(μg/ml),穿透率由19%降到5%(P〈0.01),两者比较有显著的统计学差异。中药CQCQD干预后,大鼠胰腺组织的病理评分示炎症和水肿改变较坩伊组减轻,干湿重比值下降(P〈0.001);CQCQD组大鼠胰腺组织的CTX浓度升高到6.4±1.7(μg/ml),穿透率升至7%(P〈0.05),与ANP组比较有统计学差异。结论:中药CQCQD能增加大鼠CTX的胰腺穿透率和局部组织药物浓度,其具体机制尚待进一步研究。
Objectives: To investigate the impact and mecimnism of ChaiQinChengQi Decoction (CQCQD) on Pancreatic Tissue Concentrations of Autibiotics in 1Lats with Acute Necrotising Pacreatitis. Methods: Eighteen male Sprague-Dawley rats randomly were divided into control group, ANP group and CQCQD - treated group ( n = 3 × 6) .ANP group was induced by 3.5% lml/kg sodium tanrochotate retrograde injection into hepatopancreatic duct. Six hours 'after induction of pancreatitis, all animals were introperitoneally injected with 420mg/Ego of CTX at 12 hours intervals for continuous 72 horn's. During this period, the rats in the COCQD group were intragastrically administered with 20 ml/kg of CQCQD at 8 hours intervals after induction of ANP. Fifteen minutes after the last intaperitoneal injection CTX on the 72 hours, blood and the pancreas samples were collected for detection of CTX concentration in serum and pancreatic tissue respectively, using high-pressure liquid chromatography. Pathologic changes and wet: dry ratio were obscrved in pancreatic tissue samples. Results: There were no significant differences in the serum concentrations of CIN in three groups. Compared to control group, ANP group trod significantly lower pancreatic tissue concentration of CTX ( 18.6 ±1.7 vs 4.4 ± 0.6 μg/ml, P 〈 0.0001 ) and CTX penetration ratio (19% VS 5%, P 〈 0.001) .Compared to ANP group, COCQD treatment in ANP rats significantly increased pancreatic tissue concentration of CYX (6.4 ± 1.7μg/nd vs 4.4 ± 0.6μg/ml, P 〈 0.05) and penetration ratio of CTX and (7% vs 5%, P 〈 0.05) with significantly decreases edema and inflammatory infiltration scores in pathologic observation as well as reduced wet: dry ratio. Conclusions: CQCQD has an elevatory effect on pancreatic tissue concentration and penetration rato of CTX in ANP rats. Further researches are needed to explore its mechanism.
