目的观察整合素连接激酶(ILK)在大鼠创面中的表达,探讨其在创面愈合中的作用及机制。方法健康SD大鼠20只,随机选取16只建立浅Ⅱ度烫伤模型,4只作为正常对照,烫伤后第1、5、10、15天分别随机选取4只大鼠,运用免疫组织化学检测创面中ILK的表达。7例人正常皮肤成纤维细胞,转化生长因子(TGF)-β1组给予TGF-β1刺激72h,同一细胞培养相同时间作为对照,荧光定量聚合酶链反应(PCR)检测各组ILK和纤维连接蛋白(Fn)的表达。结果烫伤后第5、10天大鼠烫伤创面中ILK的表达显著高于正常皮肤和愈合后(第15天,P〈0.05)。TGF—β1组细胞中ILK和Fn mRNA表达均显著强于对照组(P〈0.05);对照组ILK与Fn mRNA表达强度呈正相关(r=0.57)。结论ILK可能同时通过参与整合素和TGF-β1信号通路,在创面愈合过程中发挥一定作用。
Objective To explore the expression of integrin-linked kinase (ILK) in normal skin and in burned wounds of the rats, and to study its effect and mechanisms on wound healing. Methods Burns models of superficial Ⅱ° burned wounds were prepared in 16 rats. Four rats were randomly selected, and the expression of ILK was detected on the day 1,5,10 and 15 by immunohistoehemistry respectively. Normal skin from 4 SD rats served as control group. The human normal skin specimens were obtained from 8 donors who accepted skin grafting. Fibroblasts isolated from the skin specimens were cultured in Dulbecco' s modified Eagle' s medium (DMEM) supplemented with 5% fetal calf serum (FCS) and TGF-β1 (5 μg/L) in TGF-β1 group. Identical fibroblasts were cultured under the same conditions without TGF-β1 in control group. The mRNA expression levels of ILK and fibmnectin in fibroblasts at the 72nd h after treatment with TGF-β1 were detected by FQ-PCR. Results ILKs were highly expressed on the wounds during a period of 5 to 10 day as compared with normal skins and the healed wounds. The expression levels of ILK and fibronectin were significantly increased at the 72nd h after TGF-β1 treatment. Conclusion The expression of ILK was significantly increased during the course of wound healing. ILK may play an important role in wound healing. TGF-β1 can induce the expression of ILK and fibronectin in skin fibroblasts. ILK possibly promotes wound healing via the interaction between integrin and TGF-β1 pathway.
