目的观察瞬间受体势M亚基1（Transient Receptor Potential Melastatin-1,TRPM1）在舌癌组织及舌癌Tca8113细胞系中的表达情况。方法选取25例舌癌组织作为实验组,15例正常舌组织作为对照组,应用免疫组化及Western-blot的方法检测TRPM1蛋白在舌癌组织及Tca8113细胞与正常舌组织中的表达;应用RT-PCR方法检测TRPM1 mRNA在舌癌细胞与正常舌组织中的表达。结果免疫组化实验结果显示,23/25例舌癌组织中TRPM1呈强阳性表达,2/25例呈阴性表达;12/15例正常舌体组织中TRPM1呈阴性表达,3/15例呈弱阳性表达。TRPM1在舌癌与正常舌组织中的表达有显著性差异（P〈0.05）;Western-blot实验显示舌癌组织及舌癌细胞系Tca8113细胞中TRPM1蛋白的表达水平高于其在正常舌组织的表达;RT-PCR实验结果也证实TRPM1 mRNA在Tca8113细胞中的表达高于正常舌组织。结论 TRPM1与舌癌的发生可能存在相关性。

英文摘要：

Objective To investigate the expression of Transient Receptor Potential Melastatin-1（TRPM1）in the human tongue squamous cell carcinoma（TSCC）and cell line Tca8113.Methods Immunohistochemistry,reverse transcription polymerase chain reaction（RT-PCR） and Western bloting were used to survey the expression of TRPM1 in the human TSCC tissue and cell line Tca8113.Results Immunohistochemistrical result indicated high expression of TRPM1 in 23 of the 25 cases of TSCC tissue,but 12 of 15 cases in normal human tongue tissue were not expressed.The expression of TRPM1 in the TSCC tissue and in the normal tongue tissue was significantly different（P0.05）.The expression of TRPM1 in the TSCC tissue and cell line Tca8113 is higher than that in the normal tissue by Western bloting.RT-PCR showed higher expression of TRPM1 in cell line Tca8113 than that in human normal tongue tissue.Conclusions There may be a close relationship between the expression of TRPM1 and the process of the tongue squamous cell carcinoma.
