使用实验室制备的脉孢霉纤溶酶组分Ⅱ和组分Ⅲ冻干粉(均达到电泳纯),进行体外血凝块溶解试验、抗凝血实验和对小鼠一般行为的影响和毒理性试验,对不同组分的溶栓、抗凝及毒理效果进行初步研究。体外抗凝试验表明,脉孢霉纤溶酶粗酶液、组分Ⅱ、组分Ⅱ+Ⅲ具有明显的抗凝血作用,其抗凝血效果明显优于尿激酶。体外溶栓试验表明,脉孢霉粗酶液和各组分对血栓的溶解率均高于80%,明显优于对照组;脉孢霉纤溶酶粗酶液、组分Ⅱ、组分Ⅲ和组分Ⅱ+Ⅲ组溶解血栓后的液体中的红细胞分散程度较好,形态完整,尿激酶组血细胞凝集成块状,说明脉孢霉纤溶酶对细胞的损伤小,可以维持细胞的完整形态和功能,其血栓溶解效果要优于尿激酶。小鼠皮下注射试验显示脉孢霉纤溶酶无出血活性。急性毒理试验显示脉孢霉纤溶酶无明显的急性毒性,对小鼠的一般行为和协调运动无影响。
Blood clots dissolve experiments in vitro, anticoagulant experiments, behavior of mice and toxicological experiments are investigated to estimate thrombolysis, anticoagulate and toxicological effects with lyophilized powder fibrinolytic enzyme II and III from Neurospora sitophila. The results of anticoagulant experiments in vitro show that crude fibrinolytic enzyme, fibrinolytic enzyme II, fibrinolytic enzyme II and III displayed anticoagulant effect obviously and their anticoagulant effects are better than urokinase. Thrombolysis experiments in vitro show that dissolving rate of crude fibrinolytic enzyme and its components are more than 80%, which is better than that of control group. In addition, cells of these groups displayed good dispersion degree and excellent form, while blood cell of urokinase agglutinated into large cubes, which indicated that fibrinolytic enzyme has little injury of cells and could maintain the integrity of the form and function of cells. As a result; their thrombolytic effects are superior to urokinase. Fibrinolytic enzyme does not lead to bleeding after hypodermic injection. And general behavior and coordinated movements of mice are not influenced by fibrinolytic enzyme.
