【目的】利用ISSR标记分析柳州九万山野生茶树种质资源遗传多样性和亲缘关系,为柳州九万山野生茶树种质资源的保护和开发利用提供参考依据。【方法】选取10条扩增条带清晰、多态性高、重复性好的ISSR引物对93株柳州九万山野生茶树单株及6个对照茶树品种(系)的遗传多样性和亲缘关系进行分析。【结果】10条引物共扩增获得112条谱带,其中105条具有多态性,占93.75%;每条引物扩增条带数为7-16条,平均多态性带10.5条;每条引物扩增的多态性带比率为80.00%-100.00%,扩增条带大小在250~2000 bpo供试材料的相似系数介于0.23~0.82,平均为0.51。采用UPGMA法进行聚类分析,结果表明,在相似系数0.51处,供试单株共聚为十大类群,其中包含3个复合组、7个独立组,在相似系数0.53处第工类群可划分为7个亚类群,分别包括63、5、5、3、2、4和5个材料。【结论】柳州九万山野生茶树具有丰富的遗传多样性大部分单株亲缘关系较近。
[Objective]The genetic diversity and relationship of wild tea germplasm resources from Liuzhou Jiuwan Mountain were studied by using ISSR markers in order to provide references for their development,utilization and protection.[Method]The 10 ISSR primers with clear,highly polymorphic,reproducible amplified bands were selected to analyze the genetic diversity and relationship amongst 93 accessions of Liuzhou Jiuwan Mountain wild tea individuals and 6 control tea varieties(lines).[Result]A total of 112 bands were amplified,of which 105 bands were polymorphic and accounted for 93.75%;each primer could amplify out 7-16 bands with 250-2000 bp in length,the average polymorphic bands were10.5;the polymorphic band rate of each primer ranged from 80.00%to 100.00%.The similarity coefficient amongst the tested materials was from 0.23 to 0.82,with an average of 0.51.The UPGMA cluster analysis results showed that all the tested tea individuals could be clustered into 10 groups at similarity coefficient of 0.51,including 3 compound groups and7 individual groups.The I group could clustered into 7 subgroups at similarity coefficient of 0.53,which included 63,5,5,3,2,4 and 5 materials in different subgroups,respectively.[Conclusion]The wild tea germplasm resources from Liuzhou Jiuwan Mountainhaverich genetic diversity,and most of individuals present closer relationship.
