通过分析稻瘟病菌RhoGAP家族基因与几丁质合酶家族基因的表达关系,研究该家族基因对细胞壁形成的影响。利用不同生物信息学软件对RhoGAP家族进行序列分析,运用RT.PCR技术获得了该家族基因的部分eDNA序列,通过实时荧光定量技术分析RhoGAP基因与几丁质合酶基因的表达关系。结果表明,稻瘟病菌数据库中8个RhoGAP基因均含有保守的GAP结构域,且为水溶性蛋白,蛋白分子量相对较大,系统进化分析结果显示,16个RhoGTP酶激活蛋白可分为两类;不同的几丁质合成酶基因在不同RhoGAP基因突变体中的表达量不同,总的趋势是上调的。说明RhoGAP基因家族蛋白可能参与了稻瘟病菌不同阶段细胞壁的合成,本结果为进一步研究RhoGAP家族基因参与多样性的代谢途径奠定了基础。
The objective of this study was to explore the influence of Rho GTPase activating proteins (RhoGAPs) on cell wall formation from expression level between RhoGAPs and chitin synthase members. Structure characteristics and properties of RhoGAP members and expression profile between RhoGAPs and chitin synthases were analyzed by bioinformatics and Real-time PCR in Magnaporthe oryzae. Eight putative RhoGAPs in M. oryzae contain conserved GAP domain and belong to WSP (water-soluble protein) with high molecular weight. Total 16 RhoGAP proteins were systematically identified into two classes. Real-time PCR showed that chitin synthase genes had differential expression levels in different mutants of RhoGAP members. RhoGAP members may involve in the synthesis of cell wall in the fungal different development stages. Our this work are helpful for studying the regulations of diverse metabolic pathways of RhoGAP members.
