目的检测中国汉族急性白血病(AL)患者和健康受试者中CYP3A4*18和CYP3A5*3基因型分布特征,探讨CYP3A4和CYP3A5基因多态性与中国汉族AL易感性的相关性。方法病例对照研究,用聚合酶链反应(PCR)后直接测序的方法,检测中国汉族AL患者91例,健康受试者200例的CYP3A4*18B和CYP3A5*3位点的基因型分布及等位基因频率。结果中国汉族AL患者和健康受试者CYP3A4*18B和CYP3A5*3基因型分布均符合Hardy-Weinberg平衡。与健康受试者比较,AL患者CYP3A4*18B基因型及等位基因频率差异均无统计学意义(P〉0.05),CYP3A5*3基因型及等位基因分布频率差异均有统计学意义(P〈0.05)。结论 CYP3A4*18B多态性与中国汉族AL的易感性可能无关,CYP3A5*3多态性与中国汉族AL的易感性可能相关。
Objective To detect the frequency distribution patterns of CYP3A4* 18B and CYP3A5 * 3 polymorphisms in Chinese Han ethnic healthy volunteers and patients with acute leukemia,and explore the correlation between CYP3A4 and CYP3A5 genetic polymorphisms and the susceptibility of acute leukemia.Methods A case-control study was carried out to compare the distribution of genotypes and genetic frequencies of CYP3A4 * 18B and CYP3A5 * 3 genetic polymorphisms between acute leukemia(n = 91) and healthy volunteers(n = 200) with polymerase chain reaction-direct sequencing method.Results The distribution of genotypes and genetic frequencies of CYP3A4 * 18B and CYP3A5 * 3 were both in accordance with Hardy-Weinberg balances(P〈 0.05).There was no significant difference in the distribution of genotype of CYP3A4* 18B between the healthy volunteers and patients with acute leukemia(P〈 0.05).But there was a significant difference in the distribution of genotype of CYP3A5* 3 between the healthy volunteers and patients with acute leukemia(P〈 0.05).Conclusion The patients with * 18B allele of CYP3A4 might not be contributed to the susceptibility of acute leukemia,while * 3 allele of CYP3A5 might be contributed to the susceptibility of acute leukemia.
