目的观察异常黑胆质成熟剂总酚(简称总酚)与顺铂、多西他赛联用对体外培养的人宫颈癌HeLa细胞增殖和凋亡的影响。方法取对数生长期HeLa细胞接种于96孔板,加入0、50、75、100、125μg/ml总酚,然后加入0.1、1、5μg/ml顺铂或5、10、20μg/ml多西他赛,培养24、48、72 h,每孔设只加相同浓度顺铂或多西他赛的对照孔和只加培养液的对照孔。用MTT法检测细胞增殖抑制率。取对数生长期的HeLa细胞接种到培养瓶中,分别加入5μg/ml顺铂、20μg/ml多西他赛、50μg/ml总酚、5μg/ml顺铂+50μg/ml总酚、20μg/ml多西他赛+50μg/ml总酚,并设对照组,培养48 h后采用流式细胞术检测细胞凋亡率,荧光倒置显微镜下观察细胞生长情况。结果用总酚联合顺铂、多西他赛培养的HeLa细胞与相同浓度顺铂、多西他赛培养者相比,细胞增殖抑制率和凋亡率更高(P均〈0.05),形态学改变更明显。结论总酚与顺铂、多西他赛联用能显著抑制HeLa细胞的增殖,促进其凋亡。总酚与顺铂、多西他赛联用有协同作用。
Objective To investigate the effects of total phenolics from abnormal savda munziq combined with cisplatin or docetaxel on HeLa cell line of cervical cancer cultured in vitro.Methods HeLa cells were treated with different drug concentrations: total phenolics combined with cisplatin and total phenolics combined with docetaxel,we chose MTT assay to test the inhibiting effects and Flow cytometry to examine cell apoptosis rate.Microscope was used to observe the growth of HeLa cells.Results The inhibiting effect and apoptosis rate of total phenolics combined with cisplatin was greater than cisplatin(P0.05),this phenomenon also appeared in total phenolics combined with docetaxel.Conclusion Total phenolics from abnormal savda munziq combined with cisplatin or docetaxel can inhibit the growth of HeLa cells obviously,and induce apoptosis in vitro,the combinations have synergistic actions.
