Explant primary culture of the tissue, which was cut from the tail of mouse, can be used to produce skin fibroblast cells. The cells grew well in both medium M199 and DMEM, and the cells proliferations are not different between cultured in M199 and cultured in DMEM （P〉 0.05）. The epithelium cells and fibroblast cells in primary culture have different sensitivity to trypsin, so that they can be separated by trypsin under the control of time and temperature when they were passaged.
