目的:检测体外HaCaT细胞内沙眼衣原体对大环内酯类抗生素的敏感性。方法:微量稀释法检测沙眼衣原体E型标准株和34例临床株在HaCaT细胞和McCoy细胞中对红霉素、克拉霉素及阿奇霉素在体外的敏感性。同一抗生素在两种细胞的最低抑菌浓度(MIC)值进行配对t检验。结果:沙眼衣原体E型标准株在两种细胞内对红霉素、克拉霉素及阿奇霉素的MIC值一致。在HaCaT细胞和McCoy细胞中测得临床株的MIC最大值,比以往报道的MIC值均有所升高。在两种细胞中红霉素、克拉霉素及阿奇霉素对临床株的MIC值经配对t检验差异有统计学意义,且在HacaT细胞中MIC值高于McCoy细胞中的MIC值。结论:体外沙眼衣原体对大环内酯类抗生素的敏感性在不同培养细胞HaCaT细胞和McCoy细胞间存在差异。
Objective: To detect the sensitivity of Chlamydia trachomatis to macrolide in the HaCaT cells. Methods: The sensitivity of standard strain of serotype E and 34 clinical isolates of Chlamydia trachomatis to erythromycin, clarithromycin, azithromycin was determined with microdilution method in both HaCaT cells and McCoy cells in vitro. A paired t-test was used to determine the significance of MIC values in these two different cells with the same antibiotics. Results: The MIC valuess of erythromycin, clarithromycin and azithromycin against the standard strain of serotype E of Chlamydia trachomatis were the same in HaCaT cells and McCoy cells. The maximal MIC values of antibiotics against clinical isolates of Chlamydia trachomatis in both HaCaT cells and McCoy cells were higher than that previously reported. The MIC values of erythromycin, clarithromycin and azithromycin against clinical isolates of Chlamydia trachomatis in HaCaT cells were significantly different from that in Mc- Coy cells as determined by paired t-test. MIC values in HaCaT cells were higher than that in McCoy cells. Conclusion: In vitro susceptibilities of Chlamydia trachomatis to Macrolide in HaCaT cells were different from that in McCoy cells.
