目的:探讨精氨酸对体外培养的人毛乳头细胞(DPC)增殖的影响.方法:选择不同浓度(0.1、0.5、2.5、12.5及62.5 mmol/L)的精氨酸作用于体外培养的DPC 48 h,应用四甲基偶氮唑蓝(MTT)比色法、流式细胞凋亡技术,测定精氨酸对DPC增殖的影响,用PCR技术测定其对血管内皮生长因子(VEGF)、成纤维细胞生长因子(FGF)、5α-还原酶(SRD5A)、雄激素受体(AR)及雌激素受体(ER)的影响.结果:精氨酸浓度为0.1 mmol/L时,明显促进DPC增殖,与对照组比较有统计学意义;当浓度为0.5 mmol/L时,其对DPC的促增殖作用最强.当精氨酸浓度为12.5 mmol/L时,其对DPC的增殖表现为抑制作用.结论:精氨酸对DPC增殖有明显影响,适当浓度的精氨酸能显著促进DPC分泌VEGF、FGF、ER,抑制AR、SRD5A,可能是通过影响相关细胞因子的分泌从而调节毛发生长.
Objective: To study the effects of arginine on human dermal papilla cell (DPC). Methods: Five different concentrations of arginine were incubated with DPC for 48h. Then the cell proliferation and apoptosis were detected by using MTT and flow cytometry respectively. At the same time, the expressions of VEGF, FGF, SRD5A, AR and ER investigated with RT-PCR. Resuits: Arginine at the concentration of 0.1 mmol/L significantly promoted DPC proliferation, with maximum effect at 0.5 mmol/L. With this concentration it stimulated the expressions of VEGF, FGF and ER and inhibited the expressions of AR and SRDSA. These effects were dose-dependent. But it suppressed the proliferation of DPC at the concentration of 12.5 mmol/L. Conclusion: Arginine at adequate concentration can promote the proliferation of DPC by stimulating the expressions of VEGF, FGF and ER and inhibiting the expression of AR and SRD5A.
