目的:观察人毛乳头细胞聚集性生长特征。方法:将人毛乳头细胞在聚乙烯乙烯醇(EVAL)膜上以不同的密度梯度进行体外培养,观察人毛乳头细胞聚集性生长特征,与普通细胞培养板培养作对照,并对膜上培养的人毛乳头细胞进行鉴定.电镜扫描观察。结果:将传代培养的人毛乳头细胞以(80~120)×10^3个/1.1cm^2接种至铺有EVAL膜上培养时,平均3~5d后出现人毛乳头细胞的球形聚集体,对照组中人毛乳头细胞呈现平铺至多层聚集生长的特征,通过Ot一平滑肌肌动蛋白(SMA)及碱性磷酸酶对毛乳头细胞形成的球形聚集体进行免疫组化检测、电镜扫描发现其与刚分离的人头皮毛囊毛乳头形态相似。将形成的毛乳头细胞微聚体在普通培养板中重新贴壁,仍具有多层平铺生长至多层聚集生长特征。结论:在EVAL膜上培养的人毛乳头细胞具有聚集成球形生长的能力,其比在普通培养基上培养更能体现毛乳头细胞聚集性生长的特征。且形态上与刚贴壁的人毛乳头相似,为研究毛乳头细胞聚集性生长特征及为毛乳头细胞诱导毛囊再生提供了新的思路。
Objective: To observe the aggregation growth of human dermal papilla cells. Methods:Human dermal papilla cells with different concentration gradients were cultured on the ethylene-co-vinyl alcohol (EVAL) membrane,with a tissue culture polystyrene plate(TCPs)as a control, and human dermal papilla cells cultured on EVAL membrane were identified by immunohistochemistry and scanned by electron microscopy. Results: It was found that the human dermal papilla cells which at(80-120)×10^3/1.1 cm^2 cultured on EVAL membrane, after three to five days on average,formed spherical aggregates, in the control group, only showed the characteristics of multi-layer aggregation, α-SMA and alkaline phosphatase, which were molecular markers of human dermal papilla cells, they were all expressed in the spherical aggregates of dermal papilla cells. For their scanning electron microscope (SEM) and found it was similar with the dermal papilla that just separated from scalp hair follicle by morphological. When the spherical aggregates of dermal papilla cells cultured in the TCPs,these cells could be re-adherent, still able to have the characteristics of multi-storey tiled together. Conclusion:human dermal papilla cells cultured in EVAL membrane have the ability to grow together into spherical aggregates, which better reflect the aggregation growth of dermal papilla cell than cultured in general tissue culture polystyrene plate, and its shape similar to a fresh dermal papilla. It provides a new way of investigating the aggregation growth and the hair-inducing of human dermal papilla cells in vitro.
