苯丙氨酸解氨酶(PAL)是酶法生产L-苯丙氨酸的关键酶,目前主要利用红酵母PAL进行转化,但红酵母PAL稳定性较差,妨碍了其工业化的应用。该研究将交联酶聚体和印迹酶的制备方法相结合,制备出新型固定化酶-苯丙氨酸解氨酶印迹交联酶聚体,筛选出最优的底物印迹分子,并考察了该固定化酶的部分特性。结果表明,反式肉桂酸是制备印迹PAL交联酶聚体的最适底物,所制备印迹交联酶聚体的最适催化温度和pH值分别是50℃和10.5,并表现出较好的重复使用性,连续催化9个批次后,仍保持了32%的酶活保留率。
Phenylalanine ammonia lyase(PAL)is a key enzyme for production of L-phenylalanine.Currently,PAL is mainly obtained fromRhodotorula PAL However,Rhodotorula PAL exhibits poor stability,which limits its industrial application.In this study,bioimprinting cross-linked enzyme aggregates of PAL(PAL-iCLEAs)is developed by combining cross-linked enzyme aggregates technology and imprinted enzyme method.The most optimal imprinting molecule substrate is screened.Moreover,some characteristics of the PAL-iCLEAs are examined.The results show that the most suitable substrates for preparing PAL-iCLEAs is tran-cinnamic acid.The optimal temperature and pH was 50℃ and 10.5,respectively.In addition,PAL-iCLEAs shows good reusability,the recovery of PAL activity still remained 32% after reusing 9times.
