目的:探讨针刺胃经穴后提取的血清对大鼠胃黏膜细胞表皮生长因子受体(EGFR)后信使物质表达的影响。方法:60只大鼠随机分为模型血清组、胃经血清组、胆经血清组、胃经血清+PD153035组和胆经血清+PD153035组,采用水浸束缚法制作胃黏膜损伤大鼠模型,利用链霉蛋白酶消化法分离胃黏膜细胞,分别用EGFR抑制剂PD153035和血清孵育胃黏膜细胞,应用酶联免疫吸附法检测PLCγ-1活性,同位素掺入法检测PKC活性,逆转录聚合酶链反应法检测c—myc的表达水平。结果:模型血清组大鼠胃黏膜细胞PLC3γ-1,PKC和c—myc微弱表达;胃经血清组和胆经血清组大鼠胃黏膜细胞PLCγ-1,PKC和c-myc呈现较强表达,其中胃经血清组表达最高,两组相比较有显著性差异(P〈0.01):胃经血清+PD153035组和胆经血清+PD153035组大鼠胃黏膜细胞PLCγ-1,PKC和c—myc的表达较弱,胃经血清组与胃经血清+PD153035组比较有显著性差异(P〈0、01)。结论:针刺胃经穴后提取的血清能诱导大鼠胃黏膜细胞EGFR后信使物质的活化,并且存在经脉-脏腑的特异性联系.
Objective To investigate the effect of Epidermal growth factor (EGFR) signal pathway on gastric mucosa impaired after electroacupuncture in rats impaired gastric mucosa cells. Methods 60 rats were divided into 5 groups randomly: Model group, Stomach (ST) meridian group, Gallbladder (GB) meridian group, ST+PD153035 group and GB+PD153035 group. Water-immersion and restrained stress method was adopted for stress-induced gastric ulcer rat model, gastric mucosa cells were separated by using pronase digestion method. Gastric mucosa cells incubated by PD153035, EGFR inhibitor and serum; PLCγ -1's expression level on gastric mucosa cells was tested by ENZYME Linked-Immuno-Sorbent Assay (ELISA), PKC by isotopemingle method; and reverse transcription polymerase chain reaction assay (RT-PCR) was used in testing the expression level ofc-myc. Results We found the expression levels of PLCγ-1, PKC and c-myc were low in stress-induced gastric ulcer rat of Model group; In contrast, there were relatively high expression level ofPLCγ-1, PKC and c-myc in ST meridian group and GB meridian group. Among those groups which had high expression level of PLCγ-1, PKC and c-myc, the ST meridian group was the highest. There was significant difference between the ST meridian group and GB meridian group (P〈0.01), relative low expression level of PLCγ-1, PKC and c-myc was seen in ST+PD153035 group and GB+PD153035 group; In addition, there was significant difference between ST meridian group and ST+PD153035 group (P〈0.01). Conclusion The experiment suggested that there was close correlation between healing effect of electroacupuncture and activation of the EGFR singling pathway on impaired gastric mucosa cells, which was also one proof for the TCM theory"Relative particularity between Meridian and Viscera".
