目的研究HGF/C-met系统在肝纤维化发生和转归过程中的表达水平,探讨其逆转纤维化的作用机制。方法60只雄性Wistar大鼠应用硫代乙酰胺(TAA)饮用水喂养,制备肝纤维化模型。42只大鼠模型制备成功后,随机分为7组,每组6只。每隔2w处死一组大鼠,留取血清检测肝功能,并检测肝组织中羟脯氨酸含量,HE和天狼星红染色,进行组织学观察,同时应用RT-PCR技术检测HGF及其受体c-Met的表达量,以正常大鼠6只为对照。结果肝纤维化各组HGF/c-Met mRNA表达显著高于正常对照组,随着停止应用TAA时间的延长,肝功能、组织学改变、羟脯氨酸含量及HGF/c-Met的mRNA表达水平均逐渐恢复,且与停药时间呈正相关关系。结论肝损伤发生后,通过增加HGF/c-Met系统的表达水平,可促进肝细胞再生,逆转肝纤维化。
Objective To examine the expression of HGF/c-Met system in experimental hepatic fibrosis rats and investigate its effect and possible mechanism on anti-fibrosis. Methods 42 Wistar male rats of hepatic fibrosis were induced by TAA and divided into 7 groups randomly (n = 6). The rats of 1 model group were sacrificed every 2 weeks. Hepatic function of and the content of hydroxyproline (Hyp) in liver tissue were detected. HE and sirius red were used to observe histological changes of liver tissue. RT-PCR was used to measure the expression of HGF/c-Met mRNA. 6 normal rats were selected as control group. Results Expression of HGF/c-Met mRNA in the model group was higher than that in control group significantly. With the cessation of TAA, hepatic function, histological change, Hyp and the expression of HGF/c- Met mRNA were approaching to those of control group, which had positive correlation with the time of TAA cessation. Conclusions After injury of liver, the increasing expression of HGF/c-Met system can contribute to promoting hepatocyte regeneration and reverse hepatic fibrosis.
