用紫外可见分光光度计分别在500 nm和340 nm考察了由肽脂质—溴化N,N-二-十六烷基-Nα-6-三乙氧基硅基己酰基-L-甘氨酰胺((EtO)3SiC3N+C5Ala2C16)形成的阳离子脂质体——Cera-some,在还原型辅酶I(NADH)的作用下,以及在乳酸脱氢酶(LDH)和底物丙酮酸的存在下,其光密度(吸光度)随时间的变化。结果表明:随着NADH浓度的增加,Cerasome悬液的光密度(吸光度)明显增加,脂质体产生了聚集,但是聚集体不能被乳酸脱氢酶重新分散;而由溴化N,N-二-十六烷基-Nα-6-三甲胺基己酰基-L-丙氨酰胺(N+C5Ala2C16)形成的阳离子脂质体聚集体,在乳酸脱氢酶的作用下,可以被重新分散。
The over-time variation of the optical density(or absorbance) of cerasome,a cationic liposome prepared from a peptide lipid,N,N-dihexadecyl-Nα-[6-[(3-triethoxysilyl)-propyl dimethyl ammonio)hexanoyl] alaninamide bromide((EtO)3SiC3N+C5Gly2C16),under the action of a reducing coenzyme I(NADH) and in the presence of lactate dehydrogenase(LDH) and the substrate,pyruvate,was studied using the ultraviolet-visible spectrophotometer at 500 nm and 340 nm respectively.The result showed that: the aggregation of the liposome occurred as the optical density(or absorbance) of the cerasome suspension increased obviously along with the concentration of NADH,but it couldn't be redispersed by LDH;nonetheless,the aggregation of a cationic liposome prepared from N,N-dihexadecyl-Nα-[6-(trimethyl ammonio)hexanoyl]alaninamide bromide(N+C5Ala2C16) could be redispersed by LDH.
