目的:通过补骨脂素对TNFα诱导血管内皮细胞组织因子的影响,以明确补骨脂素在血栓形成过程中的作用,探讨补骨脂素对心血管系统的调节作用。方法:体外培养人脐静脉内皮细胞(human umbilical vein cells,HUVECs),采用MTT方法,检测补骨脂素对HUVECs细胞活力的影响;以肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)刺激HU-VECs,利用ELISA方法检测补骨脂对TNF-α诱导HUVECs组织因子(Tissue Factor,TF)水平的影响。结果:补骨脂素可以增强正常HUVECs活力,最大浓度约为10^-7M。在10^-7、10^-8M浓度下补骨脂素对TNF-α诱导HUVECs分泌TF呈现显著抑制作用(P〈0.01和P〈0.05)。结论:补骨脂素能够降低TNFα诱导HUVECs的TF释放,降低血栓形成风险。
Objective:The aim of this study is to investigate the effect of Psoralen on the cells viability and TF production in HUVECs,and to demonstrate the regulation on cardiovascular system by Psoralen in procedure of thrombosis.Methods:The HUVECs were successfully cultured in vitro.HUVECs were stimulated with 5ng/mL TNF-α.The influence of Psoralen on HUVECs viability was evaluated by MTT method.Furthermore,the protein levels of TF were determined by ELISA.Results:Psoralen can obviously increase the cells viability of HUVECs at the dose of 10^-7M.The TNF-α-induced TF expression was significantly decreased by pretreatment with 10^-8 or 10^-7M Psoralen.Conclusion:Psoralen has the ability to decrease the TNF-α-induced TF expression in HUVECs.Hence,it indicates that psoralen may reduce the risk of thrombotic formation through the regulating coagulation.
