目的研究Wistar母鼠在孕前和孕期喂灌十溴联苯醚(BDE-209)对子代大鼠卵巢和软骨发育的影响。方法20只雌性Wistar大鼠,随机分成观察组和对照组各10只。观察组雌鼠胃灌BDE-2091500mg/(kg·d),对照组胃灌等量的纯花生油,30d后按雌雄比例1:2合笼,雌鼠继续经口给药,方法同前,直至分娩后停止胃灌。出生后21d处死雌性子鼠,取右侧卵巢、膝关节,观察组织学改变,采用免疫组化与图像分析技术,测定卵母细胞GDF-9,软骨表层2型胶原蛋白的表达水平。结果观察组子代雌鼠卵母细胞GDF-9含量较对照组明显减少(P〈0.01);膝关节骺板软骨增厚,软骨表层2型胶原蛋白表达明显减少(P〈0.05)。结论孕前和孕期喂灌BDE-209可造成子代大鼠卵巢和软骨发育异常。
Objective To evaluate the effect of BDE -209 (brominated dipheNylethers -209) exposure before and during pregnancy on the development of ovary and articular cartilage in female rats. Methods Twenty female Wistar rats were randomly divided into two groups. Peanut oil suspensions of BDE -209 was given in a doses of 1 500 mg/( kg · d) by oral garage for the experiment group, and the controligroup was given the same volume peanut oil only. After 30 days, female rats were kept together with male rats at a proper proportion. Oral gravage was discontinued after delivery. Twenty - one days later, female neonatal rats were killed and the ovary and knee joint were obtained for HE staining and examination of the structure of the ovary and plate cartilage. Immunohistochemistry and image analysis technique were used to determine the content of GDF -9 (growth differenciation factor- 9) in the ovary and type II collagen in the superficial layer of plate cartilage. Results There was no significant difference in ovary architecture between two groups. The content of GDF- 9 in the ovary of offspring rats in experiment groups was significantly less than that of the control group (P 〈 0. 05). In the superficial layer of articular cartilage, the expression of type II collagen in the experiment group was lower than that of control group ( P 〈 0. 05 ). Conclusion Maternal BDE - 209 exposure before and during pregnancy leads to impaired development of the ovary and articular cartilage in the offspring rats.
