采用碱法提取枸杞植物叶中的蛋白质,研究了提取剂浓度、料液比、提取温度和提取时间对枸杞植物叶中蛋白质提取率的影响,确定了最佳工艺条件。正交实验结果表明,影响枸杞叶蛋白质提取率的主要因素顺序依次为:料液比>NaOH浓度>提取温度>提取时间。在NaOH浓度0.2 mol·L^-1,料液比1∶70(g/mL)和温度45℃的提取条件下提取100 min,枸杞叶蛋白质得率可达75.3%。本法简便快速、重复性好,结果准确可靠。
The protein in the leaves of Lycium chinense Mill.was extracted by alkali extraction.The effect of ex-tractant concentration,solid-liquid ratio,temperature and extraction time on the extraction rate of the protein in the leaves of Lycium chinense Mill.was studied by the orthogonal test.The optimum technological conditions were determined.The results show that the factors influencing the extraction of the protein were in the order of solid-liquid ratio>NaOH concentration>extraction temperature>extraction time.The maxmium extraction rate of the protein in the leaves of Lycium chinense Mill.is 75.3%,with the followsing extraction conditions:0.2 mol/L NaOH,solid-liquid ratio of 1∶70 (g/mL),temperature 45 ℃ and the extraction time 100 min.The method is simple,rapid,reproducible,with accurate and reliable results.
