目的:应用基因芯片技术研究经脂多糖(LPS)刺激正常人子宫内膜细胞的差异基因表达谱改变。方法:实验分LPS组与空白组,抽提空白组和LPS组中的mRNA来制备探针,经杂交、洗涤后,通过计算机扫描分析空白组和LPS组基因表达谱的差异情况。结果:与空白组比较,LPS组出现表达差异基因共290条,其中ratio≥2的上调基因181条,而ratio≤0.5的下调基因109条,涉及细胞因子、细胞周期及凋亡等相关基因。结论:LPS作用子宫内膜细胞影响了细胞内一系列基因的表达,这些基因的表达可能为从分子水平探讨炎性子宫内膜异常出血的发病机制提供基础。
Objective: To investigate the differentially expressed genes in endometrial cells stimulated by Lipoppolysacchride ( LPS ) using gene chip expression profiles.Methods: The cells were divided randomly into LPS group and blank group.Total RNA was extracted from cells.The mRNA was used to prepare probes.The mixed probes were hybridized to the gene chip expression profiles.After washing, the DNA chips were scanned by laser scanner, The acquired was analyzed. Results: 290 differentially expressed genes were found between LPS group and control group.181 genes with ratio above 2 were up-regulated, 109 genes with ratio below 0.5 were down-regulated, these genes were involved in cell factor, cell cycle and apoptosis.Conclusion: These gene expression profiles in endometrial cells induced by LPS may one of mechanisms of abnormal bleeding.
