目的:测定天山雪莲提取物的平衡溶解度及表观油水分配系数。方法:采用HPLC法测定了天山雪莲提取物在水和7种有机溶剂中的平衡溶解度,及天山雪莲提取物在正辛醇-水/缓冲盐溶液中的表观油水分配系数。结果:分别以绿原酸和芦丁为指标,天山雪莲提取物在水中的平衡溶解度为400.81mg·L-1和559.65mg·L-1表观油水分配系数为0.57(logPapp=-0.24)和0.61(logPapp=-0.22),甲醇中溶解度较大,分别为974.73mg·L-1和2413.64mg·L-1。结论:天山雪莲提取物的脂溶性较好,其平衡溶解度和油水分配系数受pH的影响较大,为天山雪莲透皮给药制剂的研究提供基础。
OBJECTIVE To determine the equilibrium solubility of Tianshan Saussurea involucrate extract and its partition coefficients for the n-octanol-water/buffer solution systems. METHODS HPLC was established and used to detect the concen tration of Tianshan Saussurea involucrate extracts in water and seven organic solvents,and also the partition coefficients for the n-octanol-water/buffer solution systems. RESULTS Chlorogenie acid and the rutin were taken respectively as the examination target, the equilibrium solubility of Tianshan Saussurea involucrate extract was 400. 81 mg.L-1 and 559. 65 mg.L-1 in water at 25℃, Papp of Tianshan Saussurea involucrate extract was 0. 57(logPapp = - 0. 24) and 0. 61 (logPapp = - 0. 22). The higher equilibrium solubility of Tianshan Saussurea involucrate extract was 974. 73 mg. L -1 and 2 413. 64 mg.L-1 in methanol. CONCLUSION The fat solubility of Tianshan Saussurea involucrata extract was good, its equilibrium solubility and the perquisite distribution coefficient were influenced greatly by the pH values, the determination may provide the foundation for the further study of the Tianshan Saussurea involucrata extracts prepartions penetrating the skin.
