目的探讨宫颈癌患者肿瘤组织中蛋白磷酸酶2A(PP2A)水平变化。方法分别选取正常对照组45例,宫颈癌患者35例。首先采用上海透镜人类乳头瘤病毒(HPV)分型试剂盒检测两组人群宫颈脱落细胞HPV感染情况;然后进一步采集宫颈组织(对照组10例,宫颈癌组8例)进行深入研究。采用酶活试剂盒检测组织PP2A酶活性,进一步采用RT-PCR技术检测PP2A在组织中的基因表达水平。结果宫颈癌患者高危型HPV感染率为94.29%,显著高于对照组(4.44%,P〈0.01);肿瘤组织中PP2A酶活性显著下调。RT-PCR检测结果发现宫颈癌组织中PP2A mRNA表达水平显著低于对照组。结论高危型HPV感染是宫颈癌发生最主要的病因。宫颈癌患者肿瘤组织PP2A酶活性和基因表达均显著下调,提示PP2A在宫颈癌的发生中起重要作用。
Objective This study aimed to discuss PP2 A levels in patients with cervical cancer tissue on the basis of the preliminary serological studies.Methods 45 cases in the control group and 35 cases in cervical cancer group were selected.Firstly we used HPV 27 types detecting assay produced by Shanghai Tellgen Life Science to detect HPV types.Then we collected cervical tissue( 10 cases of control group,8 cases of cervical cancer group) for further study.We used the enzyme assay kit to detect the tissue PP2 A activity,and used RT-PCR technology to detect PP2 A gene expression( mRNA) in tissue levels.Results Compared with the control group 4.44%,high-risk type HPV infection rate for cervical cancer patients was 94.29%( P〈 0.01).Tissue PP2 A activity decreased significantly in cervical cancer group.The expression of PP2 A mRNA in cervical cancer tissues was significantly lower than that in the control group by RT-PCR.Conclusion High-risk type HPV infection is the main cause of the occurence of cervical cancer.PP2 A activity is significantly lower in patients with cervical cancer tissue,and PP2 A mRNA expression also decreases,which indicates that PP2 A plays an important role in the occurrence of cervical cancer.
