目的利用基因芯片筛选与精子发生相关的基因,并研究其表达特征。方法将4 d、9 d、18 d、35 d、54 d和6月龄小鼠睾丸组织cDNA与Affymetrix全基因组芯片进行杂交,筛选出差异表达的基因。通过RT-PCR分析差异表达基因在小鼠睾丸不同发育阶段中的表达。结果分析Affymetrix全基因组芯片杂交结果后,筛选得到1个差异表达杂交点,通过在NCBI网站与小鼠全基因组序列Blast分析可知该差异表达基因是Rfx2基因。该基因在4 d、9 d、18 d、35d、54 d和6月龄小鼠睾丸中的芯片信号校正值分别是22.9(A)、1.5(A),130.3(P)、65.6(P)、112.7(P)和71.7(P),表明Rfx2基因在4日、9日龄小鼠睾丸中不表达,从18日龄后开始表达,而且在18日龄-6月龄小鼠睾丸中持续表达;RT-PCR结果表明Rfx2基因在4 d、9 d小鼠睾丸中不表达,在18日龄小鼠睾丸中开始表达,其实验结果与芯片分析结果一致。结论 Rfx2基因为小鼠年龄依赖性表达基因,推测Rfx2基因在小鼠精子发生过程中起重要作用。
Objective To screen the genes which were significantly changed in mouse spermatogenesis.Methods Testes cDNA samples from Balb/C mice of different postnatal days were hybridized with mouse whole genome Affymetrix chip to screen the spermatogenesis-related genes.Results The characteristics of the different-expressed genes were analyzed by various bioinformatics tools and finally one gene was chosen to furtherly investigate,which was confirmed as Rfx2 by the database in NCBI.The scaling hybridization signal intensities of Rfx2 were gradually heightened with developmental stages of mouse testis.For instence,the signal intensities on postnatal day 4,day 9,day 18,day 35,day 54,and 6 months were 22.9(Absent expression,A),1.5(A),130.3(Present expression,P),65.6(P),112.7(P) and 71.7(P),respectively.By combination with the RT-PCR analysis of mouse Rfx2,we further ensured that mouse Rfx2 begins to express from the postnatal day 18 and it is an age-dependent gene in mouse testis.Conclusion In the end,Rfx2 was considered that it would have an important role which was involved in male mammalian spermatogenesis.
