目的探讨CCK-8对RSC-364细胞株TNF受体基因表达的影响.方法采用RT-PCR法检测不同浓度的八肽胆囊收缩素(CCK-8)对在TNF-α诱导下的大鼠滑膜细胞BSC-364 TNF受体(Tumor necrosisfactor receptor,TNFR)mRNA表达的影响.结果 CCK-8在10-6mol/L和10-7mol/L浓度时可抑制TNF-α诱导的TNFR2 mRNA在RSC-364细胞的表达,且表现了一定的剂量和时间依赖性,CCK受体拮抗剂丙谷胺则可抑制此作用.结论 CCK-8可抑制TNF-α诱导的大鼠滑膜细胞RSC-364株TNFR2基因的表达.
Objective To explore the influence of choleeystokinin oetapeptide (CCK-8) on the expression of TNF receptor (TNFR) mRNA in rat synovial cells (RSC-364). Methods Detect the expression of TNFR mRNA under induction of TNF-α in the RSC-364 cells containing different concentrations of CCK-8 by RT-PCR. Results The CCK-8 at concentrations of 10^-6 mol/L and 10^-7 mol/L showed dose-and time-dependent inhibitory effect on the expression of TNFP,.2 mRNA in RSC-364 cells under induction of TNF-α,whieh could be reversed by a CCK receptor antagonist proglumide. Conclusion CCK-8 inhibited the expression of TNFR2 mRNA in RSC-364 cells significantly.
