为提高甘草及膨润土的附加值及酶的固定化寻找新载体提供一种新方法,采用新疆夏子街膨润土作基质粘土,以十六烷基三甲基溴化铵(CTAB)为模板剂,正硅酸四乙酯(TEOS)为硅源制备层柱粘土,采用吸附法固定化β-D-葡萄糖醛酸苷酶,并对该固定化酶载体进行表征和酶活测定。结果表明:层柱粘土的孔径分布在2-50nm,属于中孔吸附剂;XRD分析大量的硅溶胶进入粘土层间,其d001值为0.982nm;FT-IR分析显示酶与层柱粘土未结合的羟基发生了氢键缔合而被固定,最高固定化酶活可达1460U/g载体。
By using cetyl trimethyl ammonium bromide as the template agent,the four ethyl silicate silicon source for preparation of pillared interlayered clay(PILC) from bentonite was studied.Absorption method was used to immobilize β-D-glucuronidase using PILC as support.The character of PILC carrier and activity of immobilized β-D-glucuronidase were investigated.The results showed that the distribution of pore size of pillared clay was between 2~50nm;XRD analysis of the d001 value was 0.982 nm;FT-IR analysis showed that the enzyme was immobilized on the PILC by hydrogen bonds,the highest immobilized enzyme activity was up to 1460 U/g carrier.
