研究了不同温度及不同Ca^2+浓度提取缓冲液加0.1%胰酶(w/v)对中华绒螯蟹Eriocheirsinensis精子顶体反应发生的诱导效果。结果表明,-20、-80和-196℃处理精子不同时间和加0.1%胰酶的含Ca^2+的人工海水作精子提取缓冲液提取精子均可有效诱导精子发生顶体反应;甚至在不含Ca^2+的精子悬液中,冷冻精子也能大量发生顶体反应;而未处理的对照组和4℃处理的精子无此现象,不同的Ca^2+浓度梯度对精子顶体反应率的影响没有明显的差异性;不含Ca^2+的人工海水中的胰酶不能诱导顶体反应。
Effects of different temperatures and Ca^2+ concentration butters added with 0. 1% trypsin(w/v)on acrosome reaction (AR) induction were investigated for spermatozoa of Chinese wooly-handed crab Eriocheir sinensis. It was found that AR was induced by - 20, - 80 and --96 ℃ preservation for different time periods, as well as by artificial seawater containing Ca^2+ and 0. 1% trypsin. AR also happened profusely in spermatozoa suspensions without Ca^2+ , while the control and 4℃ treatment did not induce AR. No significant difference was found for AR induction by different Ca^2+ concentrations, and trypsin in artificial seawater without Ca^2+ did not induce AR.
