目的:探讨HIF-1α和基质金属蛋白酶10、11、13、3在乳腺癌细胞系MDA-MB-231中的表达及相关性。方法:应用RNAi和实时荧光定量PCR技术,检测乳腺癌细胞系中HIF-1α干扰前后的MMP10、11、13、3的表达变化。根据2-ΔΔCt相对定量法表示实验组相对于对照组目标基因原始拷贝数的倍数差异。结果:在HIF-1α的干扰效率是35.6%的基础上,MMP10上调了2.434倍,MMP13下调了0.448倍,差异有统计学意义(P〈0.05)。MMP11上调了0.050倍,MMP3是上调了0.257倍,差异不显著(P〉0.05)。相关分析表明,HIF-1α分别与MMP10、MMP13之间存在显著相关(r=0.943,P〈0.05),而HIF-1α与MMP11、MMP3之间的相关性不显著。结论:HIF-1α可在mRNA水平上通过对靶基因MMP10、MMP13表达水平的调节,来参与乳腺癌细胞的侵袭和转移过程。
Objective:To investigate mRNA expression of HIF-1α and matrix metallo proteinase 10,11,13 and 3 gene as well as their correlation in breast cancer cell line MDA-MB-231.Methods: miRNA interference and real-time quantitative PCR technique was used.Results: Comparing with control groups,MMP10 increased 2.434 times and MMP13 decreased 0.448 times in cells with 35.6% of interference efficiency of HIF-1α,showing a significant difference(P0.05).While the MMP11 and MMP3 increased only 0.050 and 0.257 times respectively with no significant difference(P0.05).Correlation analysis showed that HIF-1α was significantly correlated with either MMP10 or MMP13(r=.943,P0.05).Conclusion: HIF-1 might participate in the process of invasion and metastasis of breast cancer cells via modulation of expression of MMP10 and MMP13.
