目的:研究益气活血中药复方对CVB乳鼠心肌细胞感染模型NADH脱氢酶亚基1和2表达的影响,以期从基因水平揭示益气活血中药复方治疗CVB3心肌炎的作用机制,进一步证实益气活血中药复方是治疗CVB3病毒性心肌炎的有效方剂。方法:通过改良的抑制性消减杂交技术(suppression subtraetive hybridization,SSH),分离大鼠心肌细胞病毒组和益气活血中药复方给药组差异表达的基因,并通过荧光RT—PCR对上述结果进行验证。结果:SSH结果显示中药组中NADH脱氢酶亚基1和NADH脱氢酶亚基2基因的表达均比病毒组中低,这一结果通过荧光RT—PCR得到验证。结论:益气活血法能够通过调节NADH脱氢酶亚基1和2的表达而实现治疗病毒性心肌炎的目的。
Objective:To study the effects of the recipe for activating blood circulation and nourishing qi on expressions of NADH dehydrogenase subunit 1 and 2 in CVB3 infection model in rat cardiac myocytes,and to explore the treatment mechanism of the recipe for activating blood circulation and nourisbing qi on CVB3 myocarditis in gene level, and to further confirm that reci- pe for activating blood circulation and nourishing qi is a valid prescription for treatment of CVB3 myocarditis. Methods : A modified suppression subtractive hybridization (SSH) was used to isolate differentially expressed genes between CVB3 infection group and treatment group with recipe for activating blood circulation and nourishing qi. These results were further verified by fluorescence RT - PCR. Results : The results of SSH showed that gene expressions of NADH dehydrogenase subunit 1 and NADH dehydrogenase subunit 2 in treatment group were lower than those in CVB3 infection group. The results of fluorescent RT - PCR agreed with that of SSH. Conclusion : The recipe for activating blood circulation and nourishing qi can achieve the treatment of viral myocarditis by regulating the expression of NADH dehydrogenase subunitl and NADH dehydrogenase subunit 2.
