目的探讨卵巢癌中lncRNA HULC与miR-372的相互作用以及二者对p62表达的影响。方法分别在卵巢癌细胞系A2780、OVCAR3中过表达lncRNA HULC、过表达miR-372,通过RT-qPCR检测lncRNA HULC、miR-372、p62 mRNA的表达水平,Western blot检测p62蛋白表达水平,并分析三者之间的相互关系。结果卵巢癌细胞系过表达lncRNA HULC后,RT-qPCR检测到miR-372表达水平下调;而过表达miR-372,lncRNA HULC表达水平下调。过表达miR-372后,p62 mRNA和蛋白表达水平下调,而过表达lncRNA HULC后,p62mRNA和蛋白表达水平上调。结论卵巢癌中lncRNA HULC与miR-372表达具有相互抑制的关系,二者共同调控p62表达。
Objective To investigate the interaction between LncRNA HULC and miR-372, and its effects on the expression of p62 in ovarian cancer. Methods LneRNA HULC and miR-372 mimics were transfected into ovarian cancer cell lines of A2780. OVCAR3 cells respectively. The expressions of LneRNA HULC, miR-372 and p62 mRNAs were detected by RT-qPCR, Western blot was used to detect the expression of p62, then the relationship between them was analyzed. Results The expression level of miR-372 was down-regulated after overexpression of LncRNA HULC in ovarian cancer cell lines, the expression level of LncRNA HULC was down-regulated after overexpression of miR-372. The expression levels of p62 mRNA and protein were down-regulated after overexpression of miR-372, but up-regulated after overexpression of LncRNA HULC. Conclusion The expression of HULC and miR-372 has a mutual inhibitory relationship in ovarian cancer, and both of them can regulate p62 expression.
