目的复制去势家兔OPF模型,研究仙灵骨葆胶囊对家兔OPF的修复作用.方法 6月龄雌性家兔手术摘除双侧卵巢,复制家兔骨质疏松模型(OVX组),同时设假手术组(SHAM组).切除与卵巢面积相当的脂肪.卵巢切除150 d后,家兔全身及脊柱的骨密度(BMD)明显降低,说明骨质疏松模型成立,在骨质疏松的基础上进行桡骨骨折手术,36只家兔分为3组,12只/组,即假手术组(假卵巢切除+假骨折手术组+生理盐水)、OPF模型组(卵巢切除+骨折+生理盐水)及300 mg/kg仙灵骨葆胶囊治疗组(卵巢切除+骨折+仙灵骨葆胶囊).各组再分成用药60 d和120 d两个时相亚组,6只/组.分析OPF家兔全身及脊柱BMD,并结合桡骨生物力学特性、骨组织形态学观察等方法检测桡骨骨折处力学特性和第三腰椎骨微结构特征,评价仙灵骨葆胶囊家兔OPF的修复作用.结果用药60 d和120 d时,仙灵骨葆治疗组全身及脊柱BMD明显高于模型组(P〈0.01);和模型组相比,仙灵骨葆治疗组桡骨骨折处的最大载荷和能量吸收能力增加,对结构刚度则无特别明显的影响;通过仙灵骨葆胶囊治疗后,第3腰椎的骨微结构恢复.结论仙灵骨葆胶囊对家兔OPF具有较好的修复作用.
Objective The model of OPF in rabbits was duplicated to investigate the recovery effect of Xianlinggubao capsule on rabbits OPF.Me thods Female rabbits, six months old, were randomly divided into an ovariectomized(OVX) group in which the rabbits were ovariectomized, and a SHAM group in which the rabbits were removed a part of fat with the amount equal to ovaries. 150 d after ovariectomy, the body and spine bone mineral density(BMD) decreased significantly, indicating that the osteoporosis model has established. After the osteoporotic model was successfully duplicated, the surgery of radial fracture was carried out. 36 rabbits were divided into three groups each with twelve rabbits: SHAM group, OPF model group and 300 mg/kg Xianlingubao capsule group, respectively. And every of the three groups was subdivided into two time phase sub-groups each with six rabbits, based on treatment with 300 mg/kg Xianlingubao capsule for 60 d and 120 d, respectively. In OPF rabbits, body and spine BMD analysis, combined with bone biomechanical properties, bone histomorphology observation method were carried out to detect the radial mechanical properties and bone fracture micro structure characteristics, and to evaluate Xianlinggubao capsule in treating rabbit OPF. Re s ults After treatment for 60 d and120 d, BMD of Xianlingubao capsule group is obviously higher than that of model group(P〈0.01);Xianlingubao capsule group had an increased the radial maximum load and energy absorption, but had no obvious effect on structure stiffness; the bone microstructure in the third lumbar spine has been restored by Xianlingubao capsule treatment. Conclus ion Xianlinggubao capsule has a good recovery effect on rabbit OPF.
