目的观察远志皂苷元(TEN)对APP/PS1双转基因小鼠突触相关蛋白PSD-95和Shank-1表达的影响。方法 50只转基因小鼠随机分为模型组、多奈哌齐组、TEN低剂量组、TEN中剂量组、TEN高剂量组5组。同月龄C57BL/6J小鼠10只作为对照,分别给予多奈哌齐和不同剂量的TEN,对照组和模型组给予等剂量的双蒸水。3个月后,用免疫组织化学和Western blot方法检测突触相关蛋白PSD-95和Shank-1的表达。结果与对照组比较,APP/PS1双转基因小鼠海马PSD-95和Shank-1阳性细胞数明显减少,PSD-95和Shank-1蛋白表达降低;TEN各剂量组PSD-95和Shank-1阳性细胞数及蛋白表达较模型组均明显增加(P〈0.01)。结论 TEN能够增加APP/PS1双转基因小鼠PSD-95和Shank-1的表达,对突触的功能和可塑性具有改善作用。
Objective To observe the effects of tenuigenin(TEN) on the expression of synapse-related protein PSD-95 and Shank-1 in APP/PS1 double transgenic mice. Methods A total of 50 transgenic mice were randomly divided into five groups: model group, donepezil group and low-dose, medium-dose, high-dose TEN groups. 10 mice of C57BL/6J with same month age were given donepezil and TEN respectively as the controlled samples, while the control and model groups were treated with the same volume of double distilled water. After three months, the expression of synapse-related protein PSD-95 and Shank-1 were detected with immunohistochemistry and Western blot. Results Compared with the control group, the numbers of positive cells in the hippocampus of APP/PS1 double transgenic mice were reduced obviously, the expression of PSD-95 and Shank-1 were lowered; the numbers of positive cells and the expression of protein in TEN group with different doses were increased more obviously than that of the model group ( P 〈 0.01 ). Conclusion TEN can obviously increase the expression of PSD-95 and Shank-1 in APP/PS1 double transgenic mice which may play a role in ameliorating the function and plasticity of synapses.
