富勒烯(C60)对RNA逆转录的影响

目的:研究富勒烯(C60)对RNA逆转录(ReverseTranscription,RT)反应的影响。方法:以斑马鱼鱼肝mRNA为模板,在M—MLV逆转录酶作用下合成cDNA,并在RT反应体系中添加不同浓度的C60分别研究C60对M—MLV逆转录酶和RNA构象的影响。结果:100μg/mLC60可显著抑制RT反应,抑制效应与cso浓度相关,但增加M—MLV逆转录酶量可以有效逆转C60对RT的抑制作用。C60与RNA在37℃避光孵育12h后,C60可特异降解28SrRNA,同样具有浓度依赖性。结论:C60不仅可显著抑制M—MLV逆转录酶活性,而且对RNA也具有一定的损伤作用,C60具有潜在的基因毒性。

英文摘要:

Objective: To elucidate the effects of fullerene (C60) on reverse transcription (RT). Methods: Template mRNA was extracted from the liver of zebrafish, synthesised cDNA with M-MLV reverse transcriptase. Different concentrations of C60 were added into the RT reaction solu- tion, to study the impact of C60 on M-MLV reverse transcriptase and RNA conformation respectively. Results: The results showed that 100 μg/mL of C6o could significantly inhibit the RT reaction, and this effect was related to the C60 concentration. The RT inhibition of C60 could be reversed by increas- ing the amount of M-MLV reverse transcriptase in the RT reaction solution. After its co-incubation with RNA in 37 ℃ for 12 h without light, C60 can specifically degraded 28S rRNA in the concentra- tion-dependent manner. Conclusion: These results demonstrate that C60 could not only inhibit the activity of M-MLV reverse transcriptase, it also has damaging effect on the RNA. C6o may have po- tential genetics toxicity.
