利用微卫星(SSR)分子标记技术对杞柳(Salix integra Thunb)和簸箕柳(snf如suchowensis Cheng)的杂交亲本进行DNA指纹图谱分析。从496对杨树(Populus)SSR标记中筛选出23对多态性较丰富的标记,共检测到101个等位基因位点.平均有效等位基因数为2.2199。引物ORNL_312、ORNL_308、ORNL_464在指纹图谱分析中的效率较高。利用16对核心引物的指纹组合可将所有78个杞柳和19个簸箕柳杂交亲本完全区分开来,并据此绘制了杞柳和簸箕柳杂交亲本的SSR-DNA指纹图谱,同时利用9、5和7对引物分别构建了杞柳1~27、杞柳28~78及19个簸箕柳的指纹图谱。
In this paper, DNA fingerprints of Salix intagra and S. suchowensis Cheng hybrid parents were analyzed using mierosatellite(SSR) markers. The samples from S. intagra and S. suchowensis were amplified and analysized with 23 pairs of polymorphic microsatellite loci and 101 alleles screened from 496 pairs of primers from Populus. The primers ORNL_312, ORNL_308 and ORNL_464 had higher efficiency in identifying samples with fingerprint. The combination of the fingerprint from 16 core pairs of primers could be used to identify all S. intagra and S. suchowensis Cheng hybrid parents. Then the SSR - DNA fingerprint of 97 hibridparents was constructed. The fingerprints of S. intagra 1-27, S. intagra 28-78 and 19 S. suchowensis were separately made up with 9, 5 and 7 pairs of SSR primers. And this study could provide theoretical proof for mating and breeding of S. intagra and S. suchowensis.
