为探讨脑心肌炎病毒对小鼠的致病性及体内病毒分布、复制情况,选用脑心肌炎病毒VR129B株进行小鼠接种,观察其临床症状,并将死亡小鼠脑组织进行病理切片,同时利用PCR及real-time PCR方法对各组织中病毒含量进行测定。结果显示,感染小鼠出现典型的神经性临床症状,感染后第4天开始死亡,第5~9天死亡达到高峰;脑组织切片可见血管镶边现象、卫星化、噬神经细胞现象及胶质细胞结节等典型的组织病理学现象,脑、心、脾、肝及胸腺均存在病毒复制子,其中脑组织病毒含量最高。表明,小鼠对脑心肌炎病毒较为敏感,感染后出现脑炎症状和病理变化,且病毒集中在小鼠脑组织中。
To explore encephalomyocarditis virus(EMCV) pathogenicity,its distribution and replica- tion in vivo ,mice were infected with EMCV VR192B and clinical symptoms were observed. Brain patholo- gical sections of died mice were prepared,and the virus titer in the different tissues was determined by PCR and real-time PCR. The results showed that the mice showed typical clinical symptoms after infection, and death started from the 4th day post infection and got its peak in the 5th--9th day post infection. There were typical tissue pathology phenomena in brain sections, such as vascular edge phenomenon, satellite change,neuronophagia and microglial nodule. Moreover, virus replicates were found in the brain, heart, spleen, liver and thymus, especially the brain with the highest level of viruses. The results indicated that mice were sensitive to EMCV. After infection, the mice appeared encephalitis, pathological changes and viruses were mainly found in the brain.
