左金丸及类方的体外溶出行为研究

目的:研究黄连和吴茱萸不同比例组成的成药在胃肠道的溶出规律。即左金丸(6:1),甘露散(2:1),茱萸丸(1:1)和反左金丸(1:6)。方法:采用浆法,以pH=1.2的盐酸溶液作为溶出介质,利用超高效液相色谱法(ultra high per-formance liquid chromatography),测定左金丸、甘露散、茱萸丸、反左金丸中的主要活性成分小檗碱、巴马汀、药根碱在3 h内的溶出量以及不同时间点的溶出速度。结果:类方中3种生物碱在3 h内的溶出量差异显著,随着黄连比例的减少,3种生物碱的溶出量依次降低,但并没有成相同比例的减少;溶出速度差异较小,都在5 min内达到最大溶出。结论:从体外溶出角度,左金丸及类方中的吴茱萸影响了黄连生物碱的溶出。

英文摘要:

AIM:To study the dissolution of the various weight ratio of coptis to evodia fruit,including Zuojin Pill(6 : 1),Ganlu Powder(2 : 1),Zhuyu Pill(1 : 1) and Fanzuojin Pill(1 : 6),calculated as 30 gram/unit,in the gastrointestinal tract.METHODS:Paddle method was used,with hydrochloric acid(pH 1.2) as solvents,the cumulative contents of berberine,palmatine and jatrorrhizin in above pills and powder were determined with ul-tra high performance liquid chromatography in 3 h and the dissolution rates at measurement intevals.RESULTS:The dissolution amount of three alkaloids in the above pills and powder was obviously different,along with the de-crease in the weight ratio of Rhizome Coptidis,in the descenting order,berberine,palmatine and jatrorrhizin,but there was no significant difference in the dissolution velocity,and reached maximum dissolution rate in 5 min.CONCLUSION:From the viewpoint of in vitro dissolution,evodia fruit in Zuojin Pill and its similar prescription may interfere with the dissolution of alkaloids from coptis.
