β-地中海贫血是在我国南方各省高发,并显现出低hepcidin水平和铁负荷现象的一类基因缺陷型疾病。以hepcidin为研究靶点,传统的中药单体提取化合物土大黄苷和胡黄连苷Ⅱ为研究对象,研究其对hepcidin表达的调节效应。通过外源性Hamp双荧光素酶报告基因及内源性mRNA的双重验证,土大黄苷、胡黄连苷Ⅱ均表现出对hepcidin表达的刺激效应。土大黄苷和胡黄连苷Ⅱ有望成为有效缓解β-地中海贫血的潜在药物。
β-thalassemia is a high incidence of gene mutation disease,which is frequently happened in the south provinces of China.It always present low hepcidin and iron overload.We want to identify Chrysaron and Kutkin Ⅱ have the prone to induce hepcidin expression.The results conformed Chrysaron and KutkinⅡ can increase hepcidin expression by Dual-luciferase reporter assay system and qRT-PCR.It suggests Chrysaron and Kutkin Ⅱ has the potency to be a new drug to treatβ-thalassemia disease.
