以银杏(GinkgobilobaL.)雄株为材料,对银杏小孢子母细胞微管的免疫荧光观察体系进行了优化,并以优化后的方法对其减数分裂时期的微管骨架变化情况进行了研究。研究发现:利用PEG包埋可将银杏样本保存1a以上,经蛋清粘舍制片可获细胞数较多的样片,且无需酶解处理即可观察到清晰、完整的银杏小孢子母细胞微管结构。此方法的应用延长了免疫标记样本的保存时间,简化了操作步骤,并减少了传统压片法所引起的细胞分布凌乱和重复叠加等现象。
Male germ cells of Ginkgo biloba L. were used as the materials to optimize the immunofluorescence observation system of microtubule in G. biloba, and the change of microtubule was preliminarily studied by the optimized observation method during the microsporogenesis of G. biloba. Result showed that the samples of G. biloba could be preserved more than one year by PEG embedding, and the sample plates of more cells could be obtained by albumen bonding. Besides, a clear and intact structure of microtubule cytoskeleton of G. biloba could be observed without enzymolysis treatment. The application of the optimized observation method could prolong the storage life of the immunofluorescencc samples, simplify the operating procedure of immunofluorescence observation, and reduce the phenomena of the disorder distribution of ceils and superim- pose caused by the traditional squash method. Thus, it provides a convenient means for future related work.
