以十八烷基季铵盐羧甲基壳聚糖(OQCMC)、二油酰磷脂酰乙醇胺(DOPE)和3--N-N′-N′-二甲基氨基乙基-氨甲酰基胆固醇(DC-Chol)三种材料,采用逆相蒸发法(REV)成功地制备了空白阳离子脂质体(CLP).以同样方法制备二元CLP-PTX(紫杉醇)、CLP-siRNA、三元CLP-PTX-siRNA阳离子脂质复合体;并用DLS,TEM,AFM,HPLC,GC,CLSM,FCM及定量凝胶电泳法对样品性能进行表征.结果表明,所制得的阳离子脂质体球形度好、表面光滑、粒径分布均匀,平均粒径和Zeta电位分别为73.54nm和+63.1mV;PTX和siRNA的载药量和包封率分别为166.0g/mL和91.9%,siRNA转染率可达70.74%;载体中三氯甲烷残留率为5.0g/mL,远低于ICH规定的上限(60g/mL).
Blank cationic liposomes (CLP) formed from octadecyl quaternized carboxymethyl chitosan, dioleoyl-phosphatidylethanolamine and 3β-[N-(N', N'-dimethylaminoethane)-carbamoyl] cholesterol were prepared by reverse phase evaporation. CLP containing paclitaxel (PTX), siRNA and PTX-siRNA complexes were prepared in the same manner. The series was characterized by dynamic light scattering, transmission electron microscopy, atomic force microscopy, gel retardation assay, high performance liquid chromatography, gas chromatography, confocal laser scanning microscopy and flow cytometry. The CLP possessed good sphericity, well-distributed particles with a size of about 80.0 nm, Zeta potential of up to 63.1 mV, siRNA loading efficiency of up to 91.6%, siRNA transfection efficiency of up to 70.74%, residual volume of trichloromethane of 5.0 μg/mL below the minimum limit of 60μg/mL--and an entrapment rate of PTX of 166.0 μg/mL.
