目的:探讨盐酸小檗碱体外抗单纯疱疹病毒1型(HSV-1)的作用及作用机制。方法:MTT法检测盐酸小檗碱对Vero细胞的毒性作用,观察HSV-1感染Vero细胞后所致的细胞病变效应(Cytopathologic effects,CPE),实时荧光定量PCR法检测盐酸小檗碱对病毒蛋白gD的mRNA表达水平,免疫印迹(Western blot)法检测盐酸小檗碱对病毒蛋白ICP27和ICP8表达的影响。结果:盐酸小檗碱半数中毒浓度(TC50)为392.5μmol/L,病毒半数抑制浓度(IC50)为66.49μmol/L,选择性指数(SI)为5.9。实时荧光定量PCR结果显示盐酸小檗碱可降低HSV-1 gD的mRNA表达水平。Western blot显示盐酸小檗碱可明显抑制病毒蛋白ICP27和ICP8的表达。结论:盐酸小檗碱具有抗HSV-1的作用,可能与抑制病毒蛋白ICP27的表达进而影响ICP8和gD的表达有关。
Objective: To study the antiviral activity and mechanism of Berberine against herpes simplex virus 1( HSV-1) in vitro.Methods: MTT assay was adopted to evaluate the cytotoxicity of Berberine.The cytopathologic effects( CPE) that HSV-1 infected Vero cells was observed.Quantitative real-time PCR assay was conducted to determine mRNA expression of late protein g D relating to viral replication.Western blot assay researched the expression of viral proteins ICP27 and ICP8.Results: The median toxic concentration( TC50) of Berberine was 392.5μmol/L,median inhibition concentration( IC50) was 66.49 μmol/L,and selectivity index( SI) was 5.9.Quantitative real-time PCR assay showed that the expression of viral protein g D mRNA was inhibited by Berberine.Western blot revealed that the expressions of viral proteins ICP27 and ICP8 were obviously inhibited by Bererine.Conclusion: It demonstrated that Berberine has antiviral activity against HSV-1 in vitro,which was maybe related to inhibiting the expression of ICP27 protein.Subsequently,the inhibition of ICP27 protein affects the expression of ICP8 and g D.
