目的通过小干扰RNA(siRNA)干扰膜联蛋白(ANX)A1基因在甲状腺乳头状癌TPC-1细胞中的表达,探讨ANXA1对甲状腺乳头状癌细胞生物学特性的影响。方法设计并筛选使用高效的siRNA在甲状腺乳头状癌TPC-1细胞株中特异性干扰ANXA1的表达,然后用CCK8法观察ANXA1干扰后对TPC-1细胞增殖能力的影响。结果筛选的siRNA可高效沉默ANXA1在TPC-1细胞中的表达,显著抑制甲状腺乳头状癌的增殖。结论 siRNA干扰ANXA1表达,可抑制甲状腺乳头状癌的增殖,提示ANXA1可能是甲状腺乳头状癌治疗的一个重要的生物学靶标。
Objective To investigate the effects of ANXA1 on the cytobiological of papillary thyroid carcinoma cells by small RNA( siRNA) knock-down the ANXA1 expression in the papillary thyroid carcinoma TPC-1 cells.Methods The designed highly efficient siRNA was used to conduct the specific interference on ANXA1 expression in the papillary thyroid carcinoma TPC-1 cells.Then the influence on the papillary thyroid carcinoma cells proliferation was observed by CCK-8 method.Results The designed siRAN efficiently inhibited the expression of ANXA1 mRNA in papillary thyroid carcinoma cells; furthermore,markedly suppressed the proliferation of papillary thyroid carcinoma cells. Conclusions The siRNA interfering ANXA1 could inhibit the proliferation of papillary thyroid carcinoma cells.
