为研究microRNA-143(miR-143)对小鼠失神经肌肉萎缩中肌纤维的影响,通过对小鼠进行坐骨神经切断术,构建小鼠腓肠肌失神经萎缩模型,并注射miR-143模拟物或miR-143抑制剂,采用实时定量PCR检测miR-143的表达量,比较肌肉湿重和肌纤维直径,研究miR-143对小鼠失神经腓肠肌的调控作用.结果显示:注射miR-143模拟物的小鼠,其肌纤维直径明显低于对照组,而注射miR-143抑制剂的小鼠,其失神经腓肠肌组织肌纤维的直径明显高于对照组.结果说明,miR-143对小鼠失神经腓肠肌肌肉萎缩的修复起着一定调节作用.

英文摘要:

To investigate the effects of microRNA-143 (miR-143) on denervated muscle atrophy in mouse, an amyotrophia mouse model of sciatic nerve resection was established, l0 nmol miR-143 mimics or 10 nmol miR-143 inhibitors were injected to denevated gastrocnemius. Then, the expression of miR-143 was detected by qRT-PCR, the wet weight of gastrocnemius and the cross section diameter of the muscle fibers were compared between the atrophying skeletal muscle and control. The results showed that the diameter of myofiber injected with miR-i 43 mimics was significantly lower than that of control group while the diameter of myofiber injected with miR-143 inhibitors was significantly higher than that of control group. The results suggested that miR-143 plaved a role in reoairing denervated muscle atrophy.
