目的分析肺动脉高压(PAH)肺组织来源的表达谱基因芯片数据,研究差异表达基因。方法检索基因表达谱汇编数据库中GSE15197数据。用非监督性聚类、监督性分类、主成分分析的方法得到可以将PAH肺组织样本与正常对照区分开的显著差异的特征基因。用基因功能注释分析和蛋白间关联分析方法描述PAH特征基因的功能及涉及的相关信号通路。结果与正常对照相比,PAH肺组织表达显著差异特征基因有167个,功能上主要涉及蛋白的乙酰化、磷酸化等生物学术语,且存在着相互关联作用。结论 PAH差异表达特征基因及其功能注释不仅为深入研究疾病发生发展机制提供重要线索,也为有效诊断和治疗疾病提供必要理论基础。
Objective Differentially expressed genes were used to analyze the profiles of gene expression in pulmonary tissue of patients with pulmonary hypertension. Methods The profiles of gene expression were searched and downloaded from Gene Expression Omnibus(GEO)repository and GSE15197 data were used. By using unsupervised clustering,supervised classification,principal component analysis,significant difference between PAH lung tissue samples and normal controls had been recognized. The function of PAH gene and related signaling pathways were described by gene function annotation analysis and protein association analysis. Results In comparison with normal control group,there were 167 differentially expressed genes in PAH lung tissues. It was mainly involved in protein acetylation,phosphorylation and other biological terms,and there was correlation among them. Conclusion The differential expression of PAH gene and its functional annotation can not only provide important clues for further study of pathogenesis in development of the disease,but it also proves to be a theoretical basis for effective diagnosis and treatment of this disease.
